摘  要:目的探讨条件恐惧应激模型建立的可行性以及此模型的有效性。方法 64只健康雄性Wistar大鼠随机分为恐惧模型组与对照组,每组32只,采用改良的Takashi法建立条件恐惧模型。通过对大鼠恐惧行为的观察、自发活动试验的测定,动态观察各组大鼠在不同恐惧应激时期行为学方面的变化;采用放射免疫学方法测定大鼠在不同恐惧应激时期血清中皮质酮、C反应蛋白浓度的变化。结果①恐惧模型组冻结时间显著高于对照组(P<0.01),证明造模成功;②第1周两组大鼠体重无显著性差异,第2、3、4周两组大鼠体重比较有显著性差异(P<0.05);③两组大鼠自发活动比较,恐惧模型组大鼠活动性显著低于对照组(P<0.05);④恐惧模型组第1、2、3、4周血清皮质酮均显著高于对照组(P<0.05);⑤恐惧模型组第1、2、3、4周血清C反应蛋白均显著高于对照组(P<0.05)。结论条件恐惧模型动物冻结时间明显升高,且行为学改变持续时间较长。血清酶学检测显示恐惧模型动物处于较高应激水平,是较为理想可靠的恐惧动物模型。Objective To discuss the feasibility and effectiveness of conditioned fear stress model in rat.Methods The 64 male Wistar rats were randomly divided into model group and control group of fear,32,per group;by using improved Takashi method conditioned fear model was established.By the behavior,spontaneous activity test of rats after fear stimulation dynamic observition the rats'behavior at different times of fear the stress changes were investigated;Radioimmunoassay was used to detect corticosterone and C-reactive protein in serum at different times of fear stress.Results 1) Group of(fear stress) model was significantly higher than control group in the freezing time test,the difference was statistically significant(P0.01),proved it is successful model.2)The body weight of two groups in week 1 had no significantly difference;in weeks 2,3,4 the body weight of two groups were significantly different(P〈0.05).3) The spontaneous activity of two groups,the activity of(fear stress) model group was significantly lower than the control group(P〈0.05).4) In week1,2,3,4,the serum corticosterone of fear steess group were significantly higher(P〈0.05).5) In week 1,2,3,4,the C-reactive protein levels in serum of fear steess group were significantly higher(P〈0.05).Conclusion Animal model of fear condition can significantly increase freezing time,and longer duration of behavioral changes.Fear of serum enzyme tests show a higher stress level of animal models,thus it is an ideal and reliable animal model of fear.
