摘  要:目的:筛选胎兔颅骨细胞原代培养中的细胞克隆,建立成骨细胞株。方法:从胎兔颅骨组织中分离培养颅骨细胞,在此基础上,运用有限稀释法行克隆传代筛选,建立成骨细胞株,并用酶组化、免疫组化及染色体染色等方法予以鉴定。结果:原代培养的颅骨细胞来源复杂,形态多样;克隆传代筛选的细胞株,细胞形态单一,碱性磷酸酶组化染色呈强阳性,骨形态发生蛋白免疫组化染色呈强阳性,钙红染色显示具有成骨能力;染色体数目形态保持相对稳定。结论:利用有限稀释和克隆传代培养筛选建立的兔成骨细胞株,生物学性状均一,遗传学性状稳定。Objective:To screen and identify the osteoblastic strain derived from fetal rabbit calvarial bone. Methods:Routine cell isolation and culture techniques were used to screen the osteoblastic clone from fetal rabbit calvarival cells. Morphology of cells and chromsome, immunohistochemical technique, were used to identify the cell strain. Results: The morphology of primary cells cultured in vitro were diversed, suggesting a multiple origin of the cloned cells. The morphology of the cloned cells were uniform. ALP stain and BMP immunochemical stain were strong positive. Calcipoxy stain showed the cell line had osteogenetic property. Chromosome diploid and kept in steady state. Conclusion: The osteoblastic strain has the morphological, biological genetical characteristics of osteoblasts.
