摘 要:目的研究成年恒河猴和大鼠脊髓神经元及星型胶质特异标记核蛋白(NeuN)和胶质纤维酸性蛋白(GFAP)在两类动物脊髓的分布及其特异性.方法取成年恒河猴和大鼠脊髓组织,冰冻切片,用NeuN抗体和GFAP抗体行免疫组织化学SP法染色,观察NeuN和GFAP在大鼠和猴脊髓的免疫反应分布,探讨抗体标识特异性.结果猴和大鼠脊髓灰质神经元胞浆、突起和胞核均有NeuN阳性染色,但大鼠脊髓染色强于猴脊髓染色,特异性更高.GFAP在大鼠和猴脊髓星型胶质细胞均有分布,白质强于灰质,两者类似.结论识别鼠类的神经元核蛋白抗体NeuN和GFAP抗体可于灵长类脊髓染色,能有效标示灵长类脊髓神经元和星型胶质细胞.Objective To study the specific identification of NeuN and glial fibrillary acidic protein(GFAP) antibody in spinal cords of adult monkeys and rats.Methods The spinal cords of monkeys and rats were harvested,and frozen sections from spinal cords were prepared.Then immunohistochemistry staining was performed to detect the specificity and distribution of NeuN and GFAP in neurons and astrocytes from the spinal cords of adult monkeys and rats.Results NeuN positive staining neurons were detected in the spinal cords of both monkeys and rats,although the most intensive staining presented in rats’spinal cords.Comparatively,GFAP labeled astrocytes showed similar staining intensity and distribution in monkeys and rats.Conclusion NeuN and GFAP,two crucial antibodies to identify neurons and astrocytes in rodents,can be used labeled effectively the neurons and astrosytes in adult monkeys.
