摘  要:目的:本研究以HepG2.2.15(2215)细胞为细胞模型,探讨芒果苷对2215细胞内β-arrestins信号通路的影响。方法:采用免疫印迹法(Western blotting)分别检测芒果苷对2215细胞中β-arrestins总蛋白含量、β-arrestins磷酸化(p-β-arrestins)水平以及EGF干预后芒果苷对2215细胞中p-β-arrestins水平的影响。结果:在无EGF干预时,芒果苷各剂量组对2215细胞中β-arrestins总蛋白含量及p-β-arrestins水平无影响;在EGF干预后,芒果苷100μg/ml能明显抑制2215细胞中p-β-arrestins水平;芒果苷50μg/ml、25μg/ml对β-arrestins磷酸化程度无影响。结论:抑制2215细胞中p-β-arrestins水平是芒果苷抗HBV和免疫调节作用的可能机制之一。Objective: Based on HepG2.2.15(2215) cells,to investigate the effect of mangiferin on the β-arrestins signal transduction pathway in 2215 cell.Methods :The protein and phosphoralation levels of β-arrestins in 2215 cell and 2215 treated with EGF were tested with Western blotting.Results: ① Mangiferin had no effect on the protein and phosphoralation levels of β-arrestins in 2215 cell untreated with EGF;②Mangiferin 100μg/ml could obviously inhibit β-arrestins phosphoralation in 2215 cell treated with EGF,but mangiferin 50μg/ml and 25μg/ml had no effect on the phosphoralation level of β-arrestins.Conclusions: Inhibiton to the phosphoralation level of β-arrestins maybe one mechanism of mangiferin to inhibit the duplication of HBV and immunoregulation.
