摘 要:目的:观察苦参素注射液对实验性自身免疫性脑脊髓炎(EAE)大鼠一氧化氮(NO)和诱导型一氧化氮合酶(iNOS)表达的影响,探讨苦参素苦参素对EAE大鼠的防治机制。方法:采用豚鼠脊髓匀浆+完全弗氏佐剂复制EAE模型,动物随机分为正常组,模型组,苦参素高、低剂量组(150,200 mg/kg),连续给药16天,并每天对各组大鼠进行神经功能学评分、苏木素-伊红(HE)和变色酸2R-亮绿(C-2R-BG)染色观察病理改变、硝酸还原酶法测定EAE大鼠血清中NO含量的变化、免疫组织化学法和RT-PCR法分别检测脊髓中iNOS及其mRNA的表达。结果:苦参素注射液150mg/kg能明显降低EAE大鼠的神经功能学评分,减轻模型组大鼠中枢神经系统炎症浸润和脱髓鞘,减少血清中NO的含量,抑制脊髓中iNOS及其mRNA的表达;与低剂量组相比,高剂量组(200mg/kg)对NO/iNOS系统的表达并没有显著差异。结论:苦参素注射液对EAE大鼠具有防治作用,其机制可能与抑制大鼠血清和脊髓中NO和iNOS的表达有关。Objective:To investigate the possible mechanism for the prevention and treatment of EAE rats.We observed the effect of matrine injection on the expression of nitric oxide(NO)and inducible nitric oxide synthase(iNOS)in experimental autoimmune encephalomyetic(EAE)rats.Methods: The EAE models were indeced by being immuned with spinal cord extracts together with complete Freund's adjuvant.female Wistar rats were randomly divided into four groups: normal control group,EAE model group,two matrine injection treatment group(150,200 mg/kg).MAT was injected intraperitoneally daily for 16 days and clinical signs were scored daily.Histopathological analysis of spinal cord was performed by hematoxylin-eosin(HE) and chromotrope 2R-brilliant green(C-2R-BG).Nitrate reductase method was used for analyzed for NO in serum level.iNOS and iNOSmRNA in spinal cord were determined by immunostaining and RT-PCR respectively.Results:The low dose of matrine injection(150mg/kg)could significantly attenuated neurological scores,and central nervous system inflammatory infiltration and demyelination in EAE rats.Further more,it could also reduced serum level of NO,suppressed the expression of iNOS and iNOSmRNA in spinal cord;but the expression of NO and iNOS in both high(200mg/kg)and low treatment group have no significant differences.Conclusion: Matrine Injection has the effect of prevention and treat on EAE rats and the mechanism is related to inhibite the expression of NO and iNOS in rats blood and spinal cord.
