摘  要:目的:观察濒危药用植物川东獐牙菜主要提取物芒果苷与苦苷协同对HepG2人肝癌细胞的作用。方法:在体外培养的人肝癌细胞中依次加入50μg/ml川东獐牙菜水提物和乙醇不同萃取部位,在镜下观察HepG2人肝癌细胞生存状态。以芒果苷和苦苷标准品为对照,用MTT法研究正丁醇部位提取物的OD值。结果:发现正丁醇部位提取的芒果苷和苦苷的含量最高,正丁醇部位提取物作用HepG2人肝癌细胞24小时后,胞质出现空泡,细胞漂浮死亡。MTT检测其OD值呈线性递增关系,即芒果苷与苦苷混合物对肝癌细胞的杀灭作用最明显,并呈浓度依赖性。结论:濒危药用植物川东獐牙菜正丁醇部位提取物芒果苷与苦苷对体外人肝癌细胞有明显抑制和杀灭作用,其协同作用优于芒果苷或苦苷分别作用。Objective: To study the medicinal plant that the mangiferin and amarogentin was extracted from the Swertia davidi Franch effects when it was added to the HepG2 hepatic carcinoma cell in vitro.Methods: Swertia davidi Franch aqueous extract and ethanol different extract parts,inoculating 50 μg/ml them into the HepG2 hepatic carcinoma cell-line and judgmenting about the cells survival by the microscope and detected OD of MTT.Results: We discovered that the part of n-butanol extractive had the high purity mangiferin and amarogentin by HPLC.The part of n-butanol extractive made kytoplasm of hepatoma carcinoma cells appeared vacuole after 24 hours,and hepatoma carcinoma cells died.MTT examined OD curves that appeared linear increasing relations.Mangiferin and amarogentin were mixed together can inhibit and kill the obviously.There were a synergism and concentration dependent.Conclusion: The mangiferin and amarogentin were mixed together can inhibit and kill the HepG2 hepatic cells.The synergism was better than it was parted.
