摘 要:从89株临床分离鸭源大肠杆菌中选择5株氨基糖苷类高水平耐药菌,应用实时荧光定量RT-PCR方法测定其主动外排基因acrD mRNA水平,选用1株中介株、1株敏感株和大肠杆菌ATCC25922作为对照,分析其mRNA转录水平与耐药强度的相关性。结果显示,5株耐药菌的acrD mRNA相对表达量均高于中介株和敏感株,其相对表达量分别是2.00、1.96、2.22、1.84、4.39,敏感对照株为1.67,中介对照株为1.75,除4号菌株外,其余4株耐药菌相对表达量和标准菌株ATCC25922具有显著性差异;同是高度耐药的4号菌和5号菌存在较大差异,5号菌相对表达量是4号菌的2.39倍,这说明对氨基糖苷类不同耐药程度的菌株acrD基因的转录、表达存在差异,且与耐药水平呈正相关,但同时存在其他重要耐药机制介导氨基糖苷类耐药。Five aminoglycoside drug-resistance strains were selected from 89 clinical strains,to detect the expression of acrD gene by real-time RT-PCR,1 midrange drug-resistance strain,1 sensitive strain and ATCC25922 as control strains.The relative levels of acrD gene expression of drug-resistance strains was higher than the midrange drug-resistance strain,the sensitive strain and ATCC25922.The relative levels of acrD gene expression of five drug-resistance strains were 2.00,1.96,2.22,1.84 and 4.39,whereas it was 1.67 and 1.75 in susceptible strain and medium resistant strain,respectively.Remarkably,as the similar resistance level of strain NO.4 and NO.5,there is a big difference of acrD gene expression,the latter was 2.32 times of the former.The relative levels of acrD gene expression of the midrange drug-resistance strain was similar to the sensitive strain,both were higher than ATCC25922.The abover result proved that the expression of acrD gene are positive correlation with the level of aminoglycoside drug-resistant,while existing other mechanism contributes to aminoglycoside drug-resistant.
