摘  要:本实验研究了球等鞭金藻(Isochrysis galbana)8701一次性培养过程中,不同温度(10、15、20、25、30和35℃)对其叶绿素荧光参数、叶绿素相对含量及细胞密度的影响。单因子方差分析结果表明,在整个培养过程中,温度对球等鞭金藻8701的生长及各叶绿素荧光参数均有显著影响(P<0.05)。多重比较结果表明:10℃处理组的荧光参数F_v/F_m(PSⅡ的最大光能转化效率)、F_v/_o(PSⅡ的潜在活性)、ΦPSⅡ(PSⅡ的实际光能转化效率)以及rETR(相对光合电子传递效率)从接种后第1天开始就明显低于其它各组并持续到实验结束;15℃处理组的上述荧光参数从接种后第4天开始明显高于其它各组;20、25、30和35℃处理组上述参数呈现先上升后下降的趋势。25℃处理组的细胞密度和叶绿素相对含量分别从第5天和第8天开始明显高于其它处理组;10℃处理组上述参数基本没有变化;15、20、30和35℃处理组上述参数随培养时间的延长而升高。相关性分析结果表明,在整个培养过程中叶绿素相对含量与细胞密度成极显著的正相关关系。荧光参数、叶绿素相对含量以及细胞密度与温度的相关性则随着温度范围及培养天数的不同而变化。综上所述,在本实验条件下,球等鞭金藻8701生长的最适宜温度为25℃。另外,还对叶绿素荧光技术在确定微藻适温范围及筛选耐低温微藻品系中的应用进行了初步探讨。The effects of different temperatures (10℃, 15℃, 20℃, 25℃, 30℃, and 35℃) on the chlorophyll fluorescence parameters, relative chlorophyll content and cell density of Isochrysis galbana 8701 were examined in batch cultures. One-way analysis of variance indi- cated that temperature had significant effects on the growth and fluorescence parameters of I. galbana 8701 during the entire experimental perigd (P〈0.05). Multiple comparison tests showed that the parameters Fv/Fm (the maximal photochemical efficiency of PS Ⅱ), Fv/Fa (the potential activity of PS Ⅱ ),ФPS Ⅱ (the actual photochemical efficiency of PS Ⅱ ), rETR (the relative electron transport rate) under 10℃ were significantly lower than those in other treatments from day 1 after inoculation and continued to the end of the experiment, while the above fluorescence parameters under 15℃ were significantly higher than those in other treatments from day 4 after inoculation. These parameters of 20℃, 25℃, 30℃, and 35℃ treatments first increased and then dropped. The cell density and relative chlorophyll content under 25℃ were significantly higher than other treatments from day 5 and day 8 respectively. The above parameters of other treatments (15℃, 20℃, 30℃, and 35℃) increased as the culture time went on, while there was almost no change in the 10℃ treatment during the entire experiment. The analysis of correlations showed that there were significantly positive correlations between relative chlorophyll content and cell density during the entire experimental period. However the correlations between fluorescence parameters, relative chlorophyll content, cell density and temperature varied with temperature range and culture age. In summary, the optimal temperature for the growth of I. galbana 8701 was 25℃under the experimental conditions.
