摘 要:目的探讨高体积分数氧(高氧)损伤状态下,新生大鼠肺组织内血小板源性生长因子(P;;DOI:10.3969/j.issn.GF)的动态变化。方法新生Wistar大鼠320只。依据吸氧体积分数(FiO2)分为实验1组(FiO2=800 mL.L-1)、实验2组(FiO2=600 mL.L-1)、实验3组(FiO2=400 mL.L-1)及空气对照组(FiO2=210 mL.L-1)。实验第1、3、5、7、14天,无菌取其肺组织,采用免疫组织化学法检测各组肺组织P;;DOI:10.3969/j.issn.GF-A、P;;DOI:10.3969/j.issn.GF-B、P;;DOI:10.3969/j.issn.GF受体(P;;DOI:10.3969/j.issn.GFR)-α、P;;DOI:10.3969/j.issn.GFR-β的表达,MetaMorph软件测定平均光密度值,以表示肺组织P;;DOI:10.3969/j.issn.GF-A、P;;DOI:10.3969/j.issn.GF-B、P;;DOI:10.3969/j.issn.GFR-α、P;;DOI:10.3969/j.issn.GFR-β蛋白的表达水平。应用SPSS11.5软件进行统计学分析。结果实验1组及实验2组第1天肺组织P;;DOI:10.3969/j.issn.GF-B表达均低于空气对照组(Pa<0.05);实验1组第5、7天,实验2组及实验3组第7天肺组织P;;DOI:10.3969/j.issn.GF-B表达均高于空气对照组(Pa<0.05)。P;;DOI:10.3969/j.issn.GFR-β高氧刺激后持续增高,3个实验组第1天、第3天时与空气对照组比较无显著差异,第5、7、14天均明显高于空气对照组(Pa<0.05)。3个实验组P;;DOI:10.3969/j.issn.GF-A、P;;DOI:10.3969/j.issn.GFR-α与空气对照组比较差异无统计学意义。结论吸入高氧使新生大鼠肺组织P;;DOI:10.3969/j.issn.GF-B、P;;DOI:10.3969/j.issn.GFR-β表达增加,是导致早期高氧肺损伤可能途径之一。Objective To explore the dynamic changes of platelet-derived growth factor(PDGF) in lung tissue of newborn rats with hyperoxia-induced lung injury condition.Methods Three hundred and twenty newborn Wistar rats were divided into 4 groups according to different oxygen concentrations(FiO2): experimental 1 group(FiO2=800 mL·L-1),experimental 2 group(FiO2=600 mL·L-1),experimental 3 group(FiO2=400 mL·L-1) and room-air control group(FiO2=210 mL·L-1).Rats in each group were killed on the 1st,3rd,5th,7th and 14th day respectively during the experiment.The expressions of PDGF-A,PDGF-B,platelet-derived growth factor receptor(PDGFR)-α and PDGFR-β in the lung tissue were observed by means of immunohistochemistry.SPSS 11.5 software was used to analyze the data.Results The expressions of PDGF-B in the lung tissue in experimental 1 group and experimental 2 group were lower than those in the room-air control group on the 1st day(Pa0.05).The expressions of PDGF-B in the lung tissue in experimental 1 group on the 5th,7th day,and those in experimental 2 group and experimental 3 group on the 7th day,were higher than those in room-air control group(Pa0.05).The expressions of PDGFR-β in lung tissue in 3 experimental groups continued to increase after oxygen exposure.And expressions of PDGFR-β in the lung tissue in 3 experimental groups had no difference with those in control group on the 1st,3rd day,but they were significantly higher than those in room-air control group on the 5th,7th and 14th day(Pa0.05).The expressions of PDGF-A and PDGFR-α in 3 experimental groups had no difference compared with those in room-air control group.Conclusion The increment of PDGF-B and PDGFR-β expressions in the lung tissues of newborn rats after oxygen exposure can be one of the causes of hyperoxia-induced lung injury.
