摘 要:目的:建立高效液相色谱法(HPLC)测定间歇性高容量血液滤过(PHVHF)患者血清、尿液及置换液中莫西沙星浓度。方法:PHVHF联合莫西沙星治疗肾衰合并感染患者8例,测定肾功、电解质。采用依利特SinoChrom ODS-BP色谱柱(5μm,4.6mm×150mm)测定莫西沙星浓度,流动相为乙腈:(10mmol/L磷酸二氢钾、1.5mmol/L四丁基硫酸氢胺)=25:75(v/v),柱温40℃,检测波长290nm,流速1mL/min。结果:血BUN及SCr、血清K+水平显著下降(P<0.05)。血清莫西沙星在0.1~10.0mg/L范围的线性回归方程为:y=4.49x-0.257,r=0.99986。准确度为98.7%~101%,日内和日间RSD为1.12%~5.7%。置换液在0.1~10.0mg/L范围的线性回归方程为:y=3.877x-0.249,r=0.9985。准确度为85.7%~95.7%,日内和日间RSD为1.34%~8.9%。尿液在0.1~10.0mg/L范围的线性回归方程为:y=3.565x-0.270,r=0.9868。准确度为97.4%~101.7%,日内和日间RSD为1.51%~11.6%。结论:HPLC方法检测血滤患者血清、尿液及置换液中莫西沙星浓度,方法简便、快速、重复性好,可满足临床研究血滤时莫西沙星药代动力学变化的要求。Objective To determine the concentration of moxifloxacin in human's serum, urine and displacement liquid with pulse high volume hemofihration (PHVHF) by high performance liquid chromatography (HPLC). Methods HPLC analysis was carried out by using yilite hypersil ODS-BP column (5 μm, 4.6 mm × 150mm) ;The flow rate was 1.0 mL/min; The detecting wave length was at 290 nm. Results Among 8 cases, the average of BUN, Scr, K+ were significantly decreased (P 〈 0.05). The linearity of calibrated curves of serum sample was in the range of 0.1 - 10.0 mg/L r = 0.999 86,w = 1/C2. The method ological recovery of moxifloxacin was 98.7% - 101%. The within day RSDs and inter-day RSDs of were in the range of 1.12% - 5.7% of drug concentration in serum. The linearity of calibrated curves of displacement liquid sample was in the range of 0.1 - 10.0 mg/L r = 0.998 5. The method ological recovery of moxifloxacin was 85.7% - 95.7%. The within day RSDs and inter-day RSDs of were in the range of 1.34% - 8.9%. The linearity of calibrated curves of urine sample was in the range of 0.1 - 10.0 mg/L r = 0.986 8.The method ological recovery of moxifloxacin was 97.4% - 101.7%. The within day RSDs and inter-day RSDs of were in the range of 1.51% - 11.6%. Condusions This method is rapid and accurate, which can be used for the pharmacokinetic study of moxifloxacin in serum and displacement liquid samples.
