摘  要:[目的]研究重金属Cr6+对小麦幼苗根系生长和DNA含量的影响。[方法]用浓度5~100 mg/L Cr6+分别处理3和10 d龄小麦幼苗,测定生长指标,采用紫外分光光度法和电泳法测定DNA含量变化。[结果]除了Cr6+>20 mg/L时,3d龄幼苗的根数小于对照根数外,其余处理根数均增加;而根长、根系鲜重和干重、DNA含量均随着Cr6+浓度的增加而降低。3d龄幼苗比10 d龄幼苗对Cr6+更敏感。琼脂糖凝胶电泳显示,对照根尖DNA带基本完整,而Cr6+处理的DNA则发生了降解,出现拖尾现象。[结论]Cr6+处理影响了小麦幼苗根系的正常生长并导致DNA含量下降。[Objective] The effect of heavy metal-Cr6+ on the root growth of wheat seedling and the content of DNA.was analyzed.[Method] The growth index of 3——10 day-old wheat seedling,treated with Cr6+ with the concentration of 5-100 mg/L,respectively,was measured,and content of DNA was tested with the method of UV spectrophotometry and electrophoresis.[Result] Except the treatment of Cr6+ 〉 20 mg/L,in which,the number of root in 3 day-old wheat seedling was less than that of the CK,the number of root in other treatments was increased;and the length fresh weight and dry weight of root,the content of DNA were increased with the concentration-increasing of Cr6+.3 day-old seedling was more sensitive to Cr6+ than 10 day-old seedling.The analysis of agarose gel electrophoresis indicated that the DNA in root tip of CK was basic integrity and it was degraded in all of treatments’,showing its smearing.[Conclusion] The treatment of Cr6+ affected the normal root growth of wheat seedling and led the content-decreasing of DNA.
