摘 要:目的:研究银杏内酯(ginkgolides,Gin)对过氧化氢(H2O2)诱导的皮层神经元及SH-SY5Y细胞(神经母细胞瘤细胞)损伤的保护作用及其机制。方法:在培养1周的小鼠皮层神经元培养基中加入H2O2,建立神经元氧化损伤模型,用同样方法建立SH-SY5Y细胞氧化损伤模型。采用MTT比色法检测细胞活力,Western Blot技术分析亲环蛋白A的表达变化。结果:H2O2可诱导神经元和SH-SY5Y细胞损伤,使细胞活力降低,可下调亲环蛋白A的表达;Gin预处理12 h可提高细胞活力,可使亲环蛋白A表达增加。结论:Gin可抑制H2O2引起的神经元及SH-SY5Y细胞损伤,该保护作用与其上调亲环蛋白A的表达有关。Objective: To study the protective effects of ginkgolides(Gin) in primary cultured neurons and SH-SY5Y cells injuried by H2O2 and its action mechanism.Methods: Oxidative injury model of neurons and SH-SY5Y cells was induced by H2O2 in mice.The cell activity was detected by MTT,meanwhile the expression of CypA was observed by Western Blot.Results: H2O2 could both induce oxidative injury in neurons and SH-SY5Y cells,decreased the cell activity,inhibited the expression of CypA.Besides ginkgolides preconditioning could both up-regulate the cell activity and the expression of CypA.Conclusion: Ginkgolides could inhibit the injury in primary cultured neurons and SH-SY5Y cells injuried by H2O2.This protective effect may be associated with the up-regulation of the expression of CypA.
