摘  要:目的:观察带3 样蛋白( A E3)在成年大鼠心肌细胞的免疫细胞化学定位,并对其生物学意义作初步的探讨。方法:采用免疫细胞(组织)化学酶染色法及免疫荧光技术研究带3 样蛋白在心肌细胞的定位,第一抗体为红细胞带3 蛋白单克隆抗体。结果:免疫化学酶染色显示带3 样蛋白存在于成年大鼠心肌细胞膜及膜的皱褶或折入处,免疫荧光法显示带3 蛋白单克隆抗体标记了心肌细胞膜和肌纤维横纹结构的部位。结论:阴离子交换蛋白( A E)免疫细胞化学定位表明该蛋白分布于心肌细胞膜及肌纤维横纹结构的部位,这种分布特性提示 A E在精确调节细胞内p H、神经体液刺激的信号传导及将收缩蛋白和细胞骨架“铆定”到胞膜的过程中起着非常重要的作用。Objective: To investigate the immunocytochemical localization of band 3 like proteins in adult rat myocardial cells and explore its biological significance. Methods: The localization of band 3 like proteins in the myocardial cells was determined with immunocytochemical enzyme staining and immunofluorescence in the adult rats. Monoclonal antibody against erythroid band 3 proteins was used as the primary antibody. Results: Enzyme staining revealed the presence of band 3 like proteins in the sarcolemma of cultured adult rat myocardial cells and invaginations into the cells. Immunofluorescence demonstrated that the membrane and transversal riblike structures in the myocardial cells were labeled by the monoclonal antibody. Conclusion: The immunocytochemical localization of the band 3 like proteins indicates that the proteins are located in the cell membrane and transversal riblike structures. This characteristic of localization of the proteins suggests that the proteins may play an important role in accurately regulating intracellular pH, signal transduction of neurohormonal stimuli and fixing contractile proteins and cellular skeleton to the membrane.
