摘 要:目的:构建一个随机20个氨基酸的丝状噬菌体肽库。方法:扩增随机合成DNA模板,经限制性内切酶XhoⅠ+ SpeⅠ酶切后克隆进噬菌粒pTMB2,构建随机丝状噬菌体肽库并检验其库容及随机性。结果:完成丝状噬菌体随机肽库的构建,其库容为2.1×108,其氨基酸分布于理论频数无显著差异。结论:丝状噬菌体随机肽库被成功构建。Objective: To construct a library of M13 PIII fusion displaying peptides composed of 20 random amino acids. Methods: Random oligonucleotides encoded with 20 peptides were amplified and the amplified products after digested with Xho Ⅰ and Spe Ⅰ were cloned into plasmid pTMB2 to construct the library. Then the number of different recombinants and the randomness of the library were identified. Results: A random peptide library with 2.1×10 8 different recombinant clones was obtained. No significant differnece was found between te amino acid distribution of the peptide library and the expected frequency. Conclusion: The random peptide library has been successfully constructed.
