摘  要:目的:研究静脉桥基因的转移及表达。方法:将转染Adv5CMV/LacZ质粒的颈静脉桥移植于同一大鼠的颈总动脉,行β半乳糖苷酶活性测定和组织化学染色。结果:腺病毒载体能使外源性基因有效地转入静脉桥并获得高效的基因表达。结论:用腺病毒载体将治疗性基因导入静脉桥,通过基因高效表达来防止血栓形成、内膜增生和粥样硬化,可能是提高静脉桥近期和远期通畅率的一个有效方法。Aim:To study transfer and expression of adenovirus mediated gene in vein graft.Method:27 wister rats underwent implantation of reversed autologous jugular vein interposition grafts in the common carotid artery.Jugular veins were excised and distended with solution containing either Adv 5 CMV(control group)or Adv 5 CMV/LacZ(treatment group)for 30 minutes,then the jugular veins were reversed and interposed into the divided carotid arteries.Proximal and distal anastomoses were performed in an end to end fashion with interrupted 11 0 polypropylene sutures.Rats were killed after 7 days and 14 days,respectively.Samples of the midsection of the vein graft were used for β galactosidase activity examination and histochemical staining.Results:In control group,the vein grafts had no β galactosidase activity.In treatment group,the amount of β galactosidase expression at 7 and 14 days after vein graft implantation was 8.32×10 -3 u/g tissue and 7.65×10 -3 u/g tissue,respectively.Histochemically staining demonstrated extensive blue coloration of the area of intima and media of each vein graft in treatment group,no blue coloration was found in control group.Conclusion:Adenovirus vector could make LacZ gene hightly transfer into vein graft and achieve high level of gene expression.
