摘  要:目的 钙和维生素D(VitD) 是影响骨发育的两个重要因素,通过建立雏鸡单纯钙缺乏、VitD 缺乏的动物模型,比较两种情况下肥大软骨细胞凋亡的变化。方法 该研究在建立不同发育阶段单纯VitD、钙缺乏肉鸡雏鸡动物模型的基础上,采用Tunel 原位末端标记检测技术、流式细胞术等方法,从形态、生化及组织学等多方面对VitD缺乏、钙缺乏所致骨生长发育障碍的机制进行了深入的研究。结果 正常软骨细胞凋亡发生在终末肥大软骨细胞处,肥大软骨细胞凋亡百分率约为34.7 % 。雏鸡单纯钙缺乏对生长板软骨细胞的凋亡无明显影响,VitD缺乏时有生长板软骨细胞的凋亡的减少。结论 VitD 缺乏、钙缺乏导致骨发育异常的机制可能不同。VitD 缺乏导致的肥大软骨细胞凋亡减少可能是VitD缺乏时软骨细胞堆积和软骨层明显增厚的重要原因之一。Objective To observe the chondrocyte development disorder caused by vitamin D (VitD) and calcium deficiency Methods Chondrocyte apoptosis was documented by using the technique of in situ end labeling, flow cytometric analysis and DNA fragmentation Chondrocyte morphologic changes were observed by H E staining Results The results showed that 34 7 % of the normal hypertrophic chondrocytes were undergoing apoptosis, the percentage and the location of apoptosis between the calcium deficient group and the control group had no significant difference, but in VitD deficient group the percentage of hypertrophic chondrocytes apoptosis was significantly lower (19 77%) It was also found that there were chondrocytes architectural disarrangement in Vit D deficient group Conclusion The study demonstrated that there are different mechanism of bone growth disorder between VitD and calcium deficient chicken Significantly lower hypertrophic chondrocytes apoptosis may be the main reason of chondrocytes accumulation and cartilage thickning caused by VitD deficiency
