人参四物汤对成瘾动物胸腺T细胞亚群增殖及IL-2R的作用研究

摘  要:目的:观察吗啡对胸腺细胞CD4,CD8细胞周期增殖、IL2的影响及人参四物汤对其调节作用。方法:小剂量递增给药建立吗啡成瘾动物模型后,采用流式细胞仪分析各组小鼠胸腺CD4、CD8细胞DNA含量、增殖指数以及IL2R的情况。结果:吗啡成瘾组小鼠CD4、CD8细胞S,G2+M期和增殖指数显著低于正常组和人参四物汤组动物,而G0/G1期高于后二者;吗啡依赖小鼠IL2R表达下降,人参四物汤组动物的T细胞表面IL2R表达有明显增加。结论:吗啡使胸腺细胞DNA复制受阻导致功能细胞减少,IL2R表达下降。Objective:To observe the affect of morphine on proliferation of cyclic cells and IL 2R in mice, and effect of Ginseng SiWu decoction. Method:Kunming mice model for chronic dependence was developed by s. c. morphine, by flow cytometry the proliferation of cyclic cells and the levels of IL 2R were assay, and the activity of Ginseng SiWu decoction was investigated. Result:The G 0/G 1 phase of T subsets(CD 4,CD 8) were significantly higher and S, G 2+M phase lower, and IL 2R expression in morphine dependence mice than Ns and Ginseng SiWu group. Conclusion:Morphine inhibited the DNA copy of cell cycle of thymus lymphocyte subsets and IL 2R expression, Ginseng SiWu decoction was able to regulate it.
