摘 要:目的探讨毒死蜱对雄性大鼠生精功能和睾丸组织酶活力的影响。方法将40只21日龄SPF级雄性Wister大鼠随机分为4组,分别为对照组(1 mg/L吐温80溶液)和低剂量(2.7 mg/kg)、中剂量(5.4 mg/kg)、高剂量(12.8 mg/kg)毒死蜱染毒组,每组10只。采用经口灌胃方式进行染毒,染毒容量10 ml/kg,每天1次,每周6天,连续暴露90 d。染毒结束后,测定睾丸组织酶[山梨醇脱氢酶(SDH)、酸性磷酸酶(ACP)和乳酸脱氢酶(LDH)及其同工酶(LDH-x)]活力,并观察睾丸组织病理学改变。结果与对照组比较,高、中剂量毒死蜱染毒组雄性大鼠精子计数和精子活动度均较低,而精子畸形率较高,差异均有统计学意义(P<0.05)。且随着毒死蜱染毒剂量的升高,雄性大鼠精子计数和精子活动度呈下降趋势,精子畸形率呈上升趋势。仅高、中剂量毒死蜱染毒组雄性大鼠睾丸组织中LDH和LDH-x活力高于对照组,差异均有统计学意义(P<0.05)。各剂量毒死蜱染毒组雄性大鼠睾丸组织中ACP、SDH活力与对照组比较,差异均无统计学意义。随着毒死蜱染毒剂量的升高,部分生精小管出现萎缩,生精上皮细胞层次紊乱,管腔中有脱落的精子和细胞,成熟精子数量明显减少。结论毒死蜱可影响雄性大鼠的睾丸发育和生精功能。Objective To explore the effects of chlorpyrifos on spermatogenesis function and enzymatic activity of male rats.Methods Forty male Wistar rats,clean degree,were randomly divided into four groups,ten in each.The rats in the low-dose group(2.7mg/kg),the moderate-dose group(5.4 mg/kg)and the high-dose group(12.8 mg/kg)were treated with chlorpyrifos through gavage,once a day for 90 consecutive days,and the volume of gavage was 10 ml/kg.The control rats were administered to the equal volume redistilled water and tween 80(solution concentration is 1 mg/L).In the end of the experiment,the number and motility of sperms in the tail of epididymis,the enzymatic activity were tested.Results The number and motility of sprems were significantly decreased in the moderate-dose group and the high-dose group(P0.05).The moderate and high-dose groups showed obvious histopathological abnormality of the testis.The activity of LDH increased in the rats exposed to 5.4 and 12.8 mg/kg groups(P0.05).Conclusion Chlorpyrifos can impact the spermatogenesis function of male rats.
