摘  要:目的:探讨CD4+CD2+5调节性T细胞对糖尿病小鼠细胞活化的影响。方法:选取BALB/c小鼠,每日腹腔注射链脲佐菌素(STZ)40mg/kg,连续5天,建立1型糖尿病模型;从小鼠内眦静脉采血,检测血糖;取小鼠尿液,检测尿糖;流式细胞术检测BALB/c小鼠脾细胞悬液CD4+T细胞中CD28分子的表达水平及凋亡细胞的数量。结果:在注射STZ第2周和4周,BALB/c小鼠CD4+T细胞中的CD28水平未见明显变化;凋亡细胞的数量在注射STZ第2周明显升高(P<0.05),第4周进一步明显升高(P<0.01)。结论:CD4+CD25+调节性T细胞可能通过抑制CD28共刺激抑制BALB/c小鼠CD4+T细胞活化,从而影响细胞免疫功能。诱导免疫细胞凋亡可能是CD4+CD2+5调节性T细胞发挥抑制作用的又一表现形式。To investigate the influences of CD+4CD+25regulatory T cells on cellular activation in diabetes mellitus mice.BALB/c mice were infected by i.p.injection of STZ.Blood glucose of individual mice was monitored by the Blood glucose meter.Mice from normal and experimental groups were sacrificed on 0d,2w and 4w after injection for collection of spleen cells.Applying Flow Cytometry to quantitatively analyze expression of CD28 molecule on CD+4 T cells and the amount of apoptotic cells.Expression of CD28 molecule on CD+4 T cells remained unchanged according to the statistics.The amount of apoptotic cells increased significantly at 2w(P0.05),and it increased further at 4w(P0.01).CD+4CD+25regulatory T cells perhaps suppress activation of CD4+ T cells by means of CD28 costimulation during diabetes mellitus in BALB/c mice,accordingly influence cellular responses.Apoptosis might be a kind of pattern for immunosuppressive function of CD4+CD25+ regulatory T cells during diabetes mellitus.
