摘  要:目的:研究胰岛素对体外培养的小鼠卵巢颗粒细胞Foxo1mRNA表达的影响。方法:未成熟小鼠卵巢颗粒细胞原代培养,添加不同浓度胰岛素,实时定量PCR(RT-Q-PCR)检测颗粒细胞Foxo1mRNA的表达。结果:RT-Q-PCR证实小鼠卵巢颗粒细胞具有Foxo1mRNA表达,且10ng/m l胰岛素作用30分钟可减少Foxo1mRNA表达(P<0.05),100ng/m l的抑制作用更强(P<0.05),随着胰岛素作用时间延长,Foxo1mRNA表达更少(P<0.05)。结论:胰岛素抑制小鼠卵巢颗粒细胞Foxo1mRNA的表达,高浓度比生理浓度的抑制作用更强。Objective:To explore the influence of insulin on the expression of mice granulosa cells cultured in vitro.methods: The primary mice granulosa cells cultured in vitro were established and the cells were treated with different concentrations of insulin.The expression of Foxo1 mRNA in granulosa cells was determined by RT-Q-PCR.Results: The expression of Foxo1mRNA in mice granulosa cells was confirmed.By RT-Q-PCR.30 minutes of 10 ng/ml insulin function could cause the decrease of Foxo1 mRNA(P<0.05),with the increase of insulin to 100 ng/ml and prolonged time,the expression of Foxo1mRNA was further down-regulated(P<0.05).Conclusions: Insulin can down-regulate the expression of Foxo1 mRNA in mice granulosa cells,and the effect is stronger in high concentration than the normal physiological concentration.
