摘 要:目的:探讨复方五味子醇提液(SRC)对链脲佐菌素(STZ)诱导的小鼠糖尿病肾病(DN)的治疗作用及可能机制。方法:雄性C57BL/6小鼠24只随机分正常对照组、DN模型组和SRC治疗组,采用腹腔注射STZ法建立DN模型,收集各组血、尿及肾组织,检测血糖、体质量及18h尿白蛋白排泄率(UAER),Westernblotting检测各组肾组织E-钙黏蛋白(E-cadherin)、α-平滑肌肌动蛋白(α-SMA)和纤溶酶原活化抑制因子-1(PAI-1)水平;免疫组化检测各组肾组织纤黏蛋白(FN)和α-SMA的表达;光镜观察肾组织形态改变。结果:与模型组比较,SRC治疗组的右肾质量和UAER较低;SRC能上调E-cadherin,下调α-SMA和PAI-1的表达(P<0.05);抑制小管间质区FN、α-SMA表达;改善小管管腔扩张或小管萎缩,减轻间质区细胞外基质堆积。结论:SRC能显著改善C57BL/6小鼠的DN,阻抑肾小管上皮细胞-间充质转化(EMT)及PAI-1表达是其作用机制。Objective: To explore effects of ethanol extract of schisandra chinensis rhizoma chuanxiong common oyster shell (SRC) on nephropathy of diabetic mouse induced by streptozotocin (STZ), and the possible mechanisms thereof. Methods: Twenty-four male C57BL/6 mice were randomly divided into 3 groups:normal control group,diabetic model group and SRC-treated group.The mouse model of diabetes was induced by injection of STZ intraperitoneally. The serum level of glucose, body weight and urinary albumin excretion rate (UAER) were measured. Furthermore,the protein expression of E-cadherin, α-smooth muscle actin (α-SMA) and plasminogen activator inhibitor-1(PAI-1) in kidney tissues were determined by Western blotting. The expressions of fibronectin (FN) and α-SMA in kidney tissues were determined by immunohistochemical method. The morphological changes of renal tissue were observed under microscope. Results: Compared with the diabetic model group, there were significantly lower urine albumin excretion rate, right kidney weight and UAER in SRC group, and up-regulated expression of E-cadherin, down-regulations of α-SMA and PAI-1 as well(P 〈 0.05). Also, there were an inhibited expression of FN and α-SMA in tubulointerstitial area, ameliorated tubule extension or atrophy and reduced extracellular matrix accumulation in interstitial. Conclusion:SRC can ameliorate the renal fibrosis with the mechanism of suppressing epithelialmesenchymal transition (EMT) and the protein expression of PAI-1.
