摘  要:目的探讨P38信号转导通路对大鼠海马神经干细胞增殖的影响。方法体外培养大鼠海马神经干细胞,将培养细胞分为2组,对照组采用神经干细胞培养液进行培养,实验组中在神经干细胞培养液的基础上添加P38通路抑制剂SB203580,Western blot方法检测细胞中P38及磷酸化P38的表达情况,MTT法检测其对神经干细胞增殖的影响。结果神经干细胞加入SB203580后P38磷酸化水平明显低于对照组,实验组可见干细胞球逐渐增大,MTT法测定结果显示细胞增殖速度与对照组比较差异有统计学意义(P<0.05)。结论 P38信号转导通路被抑制后,神经干细胞的增殖能力明显增强。Objective To study the effects of P38 signaling pathway on the proliferation of neural stem cells (NSCs) from rat hippocampus. Methods NSCs were cultured in vitro and were divided into control and experimental groups. The control group was cultured in the NSCs media while the experimental group was cultured in NSCs media added with P38 inhibitor,SB203580. The expression of P38 and phosphorylated P38 was detected by Western blot and the proliferation of NSCs was evaluated by MTr. Results Compared with the control group, the expression of phosphorylated P38 in NSCs was significantly lower. Gradual increase of the NSC spheres was observed in the experimental group,and MTT showed that proliferation speed of NSCs was significantly faster in the experimental group ( P 〈 0.05 ). Conclusion The proliferation of neural stem cells significantly increases when P38 signal transduction pathway is inhibited.
