摘 要:目的探讨大肠癌早期肝转移的相关基因,为大肠癌肝转移的早期诊断提供理论依据。方法应用RT2 ProfilerTM PCRArray Human Tumor Metastasis(PAHS-028A)芯片,对原发灶和肝转移灶组织进行差异基因筛选。结果我们发现以下基因在大肠癌原发灶中表达明显高于肝转移结节:ACTB、APC、CTNNA1、NR4A3、MMP10、CTSL1、RB1、HPSE、ETV4、GNRH1、CDKN2A、KISS1R、IL8RB、ITGA7、ITGB3、DENR、RPSA,CXCR4、MYCL1、NME2、PNN、SMAD2,4、MMP11、SRC、RORB、SSTR2、SYK、TCF20、MMP3、TIMP2,3,4、TIPM1;以下基因肝转移灶表达明显高于原发灶:MMP9、FN1、CST7、CCL7、MGAT5。结论筛查大肠癌早期肝转移相关基因可以为合成大肠癌早期肝转移基因芯片提供依据。Objective To explore genes related to early-stage hver metastasis from colorectal cancer and provide the theoretical basis for the early diagnosis of liver metas 'tasis from colorectal cancer. Methods The RT2 ProfilerTM PCR Array Human Tumor Metastasis ( PAHS- 028A ) chip was used to screen differential genes in the primary tumor and the liver metastases. Results It was found that the following genes were expressed at significantly higher levels in the primat7 tumors of colorectal cancer than in the hver me,static nodules:ACTB, APC , CTNNA 1, NR4A3 , MMP10, CTSL 1, RB1 , HPSE , ETV4 , GNRH I , CDKN2A , KISS 1 R, IL8B B , ITGA 7,1TGB3, DENR , RPSA , CXCR4 , MYCL1 , NME2 , PNN , SMAD2, 4 , MMPl l , SRC , RORB , SSTR2 , SYK , TCF20 , MMP3 , TIMP2 , 3 , 4 and TIPM I. The tbllowing genes were expressed higher in liver metastases than in the primary tumor: MMP9, FNI, CST7, CCL7 and MGAT5. Conclusion Screening for genes related to early-stage liver metas 'tasis genes may provide basis for making gene chips of early-stage liver metastasis from colorectal cancer.
