淋巴淤滞诱发大鼠胰岛超微结构变化的透射电镜观察

摘  要:目的探讨实验性淋巴瘀滞对实验动物胰岛超微结构的影响。方法选用30只SD大鼠,随机分为对照(假手术)组10只和模型(手术)组20只。术后6个月取胰组织标本进行透射电镜样品制备和观察。结果与对照组相比,模型组大鼠的胰腺小叶和胰岛细胞间隙明显增宽,间隙内出现大量脂滴样物质、红细胞和胶原原纤维样结构;可见分泌颗粒大小和电子密度不同的胰岛细胞,在胰岛B细胞内,有些分泌颗粒的晕环明显增宽;有些胰岛细胞的核,其核膜变的模糊不清,出现核染色质边集甚至核固缩现象。结论胸导管结扎可引起胰淋巴回流障碍,诱发胰岛细胞的超微结构变化。Objective To explore the influence of the ultra-structural changes of pancreatic islet induced by experimental lymphostasis.Methods 30 SD rats were divided into 2 groups randomly model operative group(20) and control(sham operative) group(10).At 6 months after operation,the ultra-sections of the pancreatic tissue was prepared and the ultra-structure was observed with transmission electron microscope(TeM).Results TeM observation displayed that the pancreatic interlobullar space and intercellar space of pancreatic islets were widened,while blood vessels and enlarged lymphatic vessels were visible;a lot of lipid droplets,erythrocyte and some collagen fibril structures could be seen.The pancreatic islet cells contained with various secretory granules of size and electron density could be seen.Within B cell of pancreatic islets,secretory granules were larger,halo rings were wider obviously,nucleus chromosome were margination,nucleus membrane became unclear,even displayed the pyknosis of nucleus.Conclusion The ligaturing rat thoracic duct can contribute to the dysfunctions of pancreatic lymph drainage,the lymphostasis lead to the ultra-structural changes of islet cells and affect the function of pancreatic islets.
