摘 要:经紫外诱变,筛选出在碳酸氢铵浓度为400 mg/L条件下可以生存的突变株。通过培养基中氮、磷、铁、温度、光照及pH值对雨生红球藻诱变株生长的影响进行比较,对以上3种营养盐及3种环境因子分别进行三因素三水平正交实验,以优化培养条件。进而对雨生红球藻野生型及诱变株的生长状况及色素含量进行比较。实验结果表明,雨生红球藻抗铵品系在培养基中NaNO30.1 g/L,KH2PO40.01 g/L,FeCl3.6H2O 1.0 mg/L,在pH值为8.0,光照强度100μmol.m-2.s-1,温度为18℃的培养条件下,雨生红球藻的生物量得到有效提高。A Haematococcus pluvialis strain that can stands in 400 mg/L NH4HCO3 was obtained by UV mutagenesis and selection.In order to determine the optimal cultural conditions,effects of nutrition elements(nitrate,phosphate and ferric) and other ecological elements(pH value,temperature and light intensity) on the growth of Heamatococcus pluvialis of mutant strains,and orthogonal tests with three elements and three levels were conducted.Also,the cell growth and photosynthetic pigment content of wild type and mutant strain were compared.Results uncovered that the optimal culture conditions of mutant strain were determined as follows: NaNO3,KH2PO4 and FeCl3·6H2O concentrations 0.1 g/L,0.01 g/L and 0.001 g/L,respectively,and pH8.0,light intensity 100 μmol·m-2·s-1and temperature 18 ℃.Under these conditions,the biomass of Heamatococcus pluvialis increased remarkably.
