摘  要:采用微囊化牛嗜铬细胞(BCC)异种移植于大鼠和癌痛病人脊髓蛛网膜下方法,观察BCC镇痛作用和海藻酸钠-聚赖氨酸-海藻酸钠(APA)微囊对异种移植物的免疫保护作用。结果显示,移植后微囊组和非微囊组大鼠基础热痛阈和尼古丁激发热痛阈明显升高;微囊组80%受试鼠的镇痛作用持续时间超过270天。将微囊化BCC植入8例癌痛病人脊髓蛛网膜下,除1例镇痛效果不明显外7例缓解,减少或停止使用止痛药,镇痛作用持续时间>60天。表明BCC异种移植于脊髓蛛网膜下可产生明显的镇痛作用,APA微囊具有良好的免疫隔离作用。To observe analgesit effects of xenogeneic bovine chromaffin cells(BCC) and protective effects of the alginate polylysine alginate (APA) microcapsule. BCC was implanted into the spinal subarachnoid space of normal rats and eight patients suffering from terminal cancer pain. The results showed that, baseline pain sensitivity and pain sensitivity following a low dose of nicotine decreased remarkly in rats after being transplanted with microencapsulated BCC and non microencapsulated BCC. In 80% of rals with transplartation of microencapsulated BCC induced analgesia was well maintained over 270 days. After transplantation of microencapsulated BCC, pain in seven patients was ameliorated, and no further analgesic drug was necessury, or the dosage of analgesic drug was reduced. The analgesia was maintained over 60 days. No obvious side effects were observed.The studies indicated BCC xenograft in the spinal subarachnoid space could rapidly produce obvious antinociceptive effects, and microencapsulation provided good immunoisolation effect.
