摘 要:目的和方法:采用两种体外细胞趋化游走实验体系及间接免疫荧光等方法,观察丹参单体(IH764-3)对rhIL-8诱导的人嗜中性白细胞(neutrophils,Neu)跨聚碳酸酯(PC)膜和人脐静脉内皮细胞(humanumbilicalveinendothelialcells,HUVEC)单层趋化游走过程的影响,以分析IH764-3的抗炎作用机制。结果:与对照组比较、IH764-3对rhIL-8诱导的Neu跨PC膜趋化游走(P<0.01)和跨HUVEC单层趋化游走(P<0.01)均有明显地抑制作用。这种抑制作用是IH764-3抗炎作用机制之一,与对照组比较、IH764-3对rhIL-8诱导NeuCD11b表达(P<0.01)及Neu与HUVEC单层粘附(P<0.01)均有明显抑制作用。结论:这种抑制作用是IH764-3抑制Neu趋化游走的深层机制。AIM and METHODS:Two in vitro cells migration test (a 48 well micro chemotaxis chamber system and a Transwell coculture system) and indirect immunofluorenscence test were used to gain a deep insight into the effects of IH764 -3 (profocatechuic aldehyde), one of monomers extracted from Radix Salviae Miltiorrhizae, a Chinese plant, on transpolycarbonate membrane and trans-human umbilical vein endothelial cells (HUVEC) monolayer chemotactic migration of human neutrophils (Neu) induced by human recombinant interleukin - 8(rhIL - 8); to analyse the mechanisms of IH64-3 antiinflammatory action. RESULTS: Compared with control group, IH764 - 3 inhibits significantly both transpolycarbonate membrane migration of Neu(P < 0.001 ) and trans -HUVEC monolayer migration of Neu (P < 0.01) induced by rhIL - 8. The inhibition is thought to be one of the mechanisms for IH764 - 3 antiinflammatory action. On the other hand, compared with control conditions, IH764-3 inhibits both CD11b expression on Neu induced hy rhIL - 8(P < 0.01 ) and rhIL-8 induced Neu adhesion to HUVEC monolayer (P <0.01 ) significantly. CONCLUSION: The inhibition is thought to be an underlying mechanisms for IH764- 3 antiinflammatory action.
