摘  要:稻纵卷叶螟等鳞翅目害虫对cry2A*编码的Bt毒性蛋白有明显的排斥效应,从而可达到抗虫的效果。本研究利用农杆菌介导法,将载有cry2A*基因的pC-2A质粒转入受体水稻品种镇稻88基因组内,获得转基因植株。由于cry2A*与抗Basta基因紧密连锁,通过Basta抗性筛选,进而得到转cry2A*基因阳性株。PCR和AGE分析结果表明,外源基因cry2A*已成功整合到镇稻88基因组内。田间试验结果显示转基因阳性植株有明显的抗虫效果。Lepidopteran pests,especially stemborers and leaffolders,had obvious repulsion to Bacillus thuringiensis(BT) toxic protein encoded by cry2A* gene,so the pC-2A plasmid with cry2A* gene was transferred into the genome of rice variety Zhendao 88 by Agrobacterium-mediated method.Then the transgenic rice plants were obtained through Basta screening based on the tight linkage of cry2A* gene with Basta-resistant gene.The PCR and AGE analysis indicated that the cry2A* gene had been successfully integrated into Zhendao 88 genome.The field test showed that the transgenic plants had much better insect resistance compared with common plants.
