摘  要:目的研究大孔树脂吸附法分离纯化橙皮中橙皮苷的最佳工艺条件,通过熔点、橙皮苷的薄层(TLC)、UV及IR图谱进行结构表征。方法以橙皮苷的纯度为指标,比较HPD826、HPD417、AB-8、HPD722及HPD600大孔树脂对橙皮苷的吸附和解吸附性能。结果 AB-8大孔树脂对橙皮苷的吸附和解吸附性能较好,吸附量为0.37 mg/g,吸附率为86.2%;10%的乙醇碱溶液(V/V)洗脱,解吸率为72.7%。结论橙皮苷的最佳纯化工艺是选用AB-8大孔树脂吸附,pH12的10%乙醇碱溶液(V/V)洗脱,所得产物纯度达91.5%,同时图谱确证了橙皮苷的结构。Objective To study the optimally purified process of hesperidin extracted from Pericarpium citri reticulatae with macroporous resin and to confirm the structure of hesperidin by melting point(M.P.),TLC,UV and IR spectra data.Methods For the purity of hesperidin as a main objective,the adsorption and desorption characters of HPD826,HPD417,AB-8,HPD722 and HPD600 macroporous resins were compared.Results AB-8 had the best adsorptive property and desorption,the adsorptive content of AB-8 was 0.37 mg/g.The adsorptive and desorptive rates were 86.2% and 72.7%,respectively.The eluted solution was 10% alcoholic and alkaline water solution(pH 12,V/V).Conclusion The optimally purified process of hesperidin is using AB-8 macroporous resin,10% alcoholic and alkaline water solution(pH 12,V/V)as eluant,the purity of hesperidin 91.5%,the spectra data had confirm the structure of hesperidin.
