旋覆代赭汤及其倍用甘补方对混合性反流性食管炎模型大鼠延髓神经核团c-Fos蛋白表达的影响  被引量:9

摘  要:目的:观察旋覆代赭汤对混合性反流性食管炎模型大鼠延髓神经核团c-Fos蛋白表达的影响。方法:采用"食管十二指肠端侧吻合术"制作动物模型。110只大鼠随机分为正常对照组、模型组、西药(莫沙必利)对照组、旋覆代赭汤原方组、原方倍用甘补组,给予相应处理或用药后,取延髓组织,用免疫组化的方法测其c-Fos的表达。结果:旋覆代赭汤原方倍用甘补组能显著提高反流性食管炎模型大鼠c-Fos的表达,与正常组、假手术和模型组相比,c-Fos的表达率显著升高(P<0.01),与原方组和西药组相比,c-Fos的表达率升高(P<0.01),而原方组与西药组相比无显著性差异(P>0.05)。结论:旋覆代赭汤原方倍用甘补组可明显升高混合性反流性食管炎延髓神经核团c-Fos蛋白表达,从而反映该药物对增强中枢神经系统的迷走-迷走反射强度,能够更高程度地激活胆碱能抗炎通路,发挥其对食管炎大鼠积极的保护作用。Objective:To observe the effect of Decoction of diplo-dose of Xuan-fu Dai-zhe on c-Fos protein expression in medullary nucleus loaded by rat model of mixed reflux esophagitis. Method:The animal models were made with esophagoduodenostomy. 110 rats were randomly divided into normal control group, model group, mosa- pride control group ,treatment group of Decoction of Xuan-fu Dai-zhe, and treatment group of Decoction of Xuan-fu Dai-zhe and diplo-dose of Gan-bu. They were treated with different method. And c-Fos protein expression was detected in medullary nucleus with immunohistochemical methods. Resuh:The expression of c-Fos was obviously im- proved in treatment group of Decoction of Xuan-fu Dai-zhe and diplo-dose of Gan-bu, which was higher remarkably than that in normal control group, sham operated group and model group ( P 〈 0. 01 ), and higher than that in mosa- pride control group and treatment group of Decoction ofXuan-fu Dai-zhe (P 〈 0. 01 ). But there was not significant difference between mosapride control group and treatment group of Decoction of Xuan-fu Dai-zhe ( P 〉 0. 05 ). Conclusion :Decoction ofXuan-fu Dai-zhe and diplo-dose of Gan-bu can increase c-Fos protein expression markedly in model of mixed reflux esophagitis. It shows that the drugs can protect the rats from esophagitis through increasing vagus-vagal reflex strength in central nervous system and activating antiinflammatory pathway of bilineurin energy.
