WHBE兔对卵白蛋白诱发变应性鼻炎的敏感性研究

摘　　要：目的通过与日本大耳白兔作比较,来探究WHBE兔对卵白蛋白诱发变应性鼻炎的敏感性。方法 WH-BE兔和日本大耳白兔各20只,各随机分成模型组和正常对照组,每组10只,隔天用卵白蛋白加氢氧化铝腹腔注射作基础致敏7次后用1%卵白蛋白溶液每侧滴鼻0.1 ml激发建立变应性鼻炎模型,观察各组兔子鼻部症状并评分,末次激发后取各组动物的鼻黏膜作甲苯胺蓝染色,计数并比较肥大细胞在各组之间的差异。结果与正常对照组比,变应性鼻炎激发后WHBE兔和日本大耳白兔模型组症状明显(P<0.01),鼻黏膜肥大细胞数量增加(P<0.01);WHBE兔激发后鼻部症状评分有高于日本大耳白兔的趋势,末次激发评分显著高于日本大耳白兔(P<0.05),鼻黏膜肥大细胞数量也显著高于日本大耳兔(P<0.05)。结论 WHBE兔对卵白蛋白加氢氧化铝诱导变应性鼻炎有较强的敏感性,较适合用于变应性鼻炎的研究。Objective To compare the allergic rhinitis model of WHBE rabbit with that of Japanese white rabbit,and explore the sensitivity of WHBE rabbit to ovalbumin-induced allergic rhinitis.Methods Twenty WHBE rabbits and 20 Japanese white rabbits were randomly divided into model group and control group（n=10 in each）.The model groups were initially sensitized every other day for seven days with intraperitoneal injection of ovalbumin and aluminium hydroxide.The rabbit models of allergic rhinitis were then established after the model groups were treated with nasal drip of 1% ovalbumin solution.The nasal symptoms were observed and graded.After the last inducement the nasal mucosa samples were collected for toluidine blue stain to show the counts and density of mast cells in each group.Results Compared with control group,the symptom of allergic rhinitis was significant（P0.01） and the number of mast cells in nasal mucosa was markedly increased（P0.01） in WHBE rabbits and Japanese white rabbits model groups.The grade of the symptom of allergic rhinitis in WHBE rabbits was higher than that in Japanese white rabbits,especially after the final inducement（P0.05）,so did the number of mast cells in nasal mucosa（P0.05）.Conclusion WHBE rabbit has better system sensitivity to ovalbumin and aluminium hydroxide induced allergic rhinitis,and is suitable for the study of allergic rhinitis.
