摘 要:品种纯度不够是建立高质量果园的重要限制因素。建立在苗期鉴定纯度的分子方法,对选择高纯度幼苗建立高质量果园有重要意义。本试验用改进的CTAB法提取柑桔叶片的总DNA,并利用梯度PCR优化了RAPD反应条件,建立了RAPD-PCR分析的最佳反应体系,显著提高了柑桔RAPD分析的特异性、效率和稳定性。从21条随机引物中扩增贡柑等4个品种的DNA,筛选出一组扩增条带多态性高、稳定性好的RAPD分析引物。结果证明,RAPD分析是品种纯度鉴定研究的简单实用的方法。Molecular methods for the identification of seedling uniformity are of great importance in choosing quality seedlings before establishing citrus orchards. In this study, an improved CTAB method was used to extract total DNA from citrus leaves. Gradient PCR was used to optimize the reaction conditions. An optimized RAPI-PCR reaction system was thus established. Results showed that the specificity, efficiency and stability of RAPD reactions were significantly improved by the method. The method was used to amplify DNAs of four local citrus varieties from Zhaoqing, Guangdong Province, using 21 pairs of random primers, and a subset of primers yielded reproducible polymorphic bands. The results showed that RAPD was a simple yet useful method for the assessment of seedling uniformity.
