摘 要:大豆异黄酮是一类具有重要保健功能的酚类次级代谢物,是大豆品质的重要指标之一。提高大豆中的异黄酮含量对大豆品质的改良有重要意义。试验利用外源茉莉酸甲酯作为诱导物对田间栽培的大豆进行叶面喷施,上调大豆异黄酮合成途径提高大豆中异黄酮的含量并分析种子成熟过程中异黄酮合成途径的动态变化。研究表明,外源茉莉酸甲酯通过提高籽粒中苯丙氨酸解氨酶(PAL)的活力、前体物—色氨酸含量来提高黄豆苷元、染料木黄酮、总异黄酮的合成;在0~600μmol·L-1的喷施浓度范围内,450μmol·L-1是最适浓度。450μmol·L-1的茉莉酸甲酯处理后所收获的大豆种子中黄豆苷元的含量比对照提高了21.5μg·g-1,染料木黄酮的含量比对照提高了5.1μg·g-1,总异黄酮的含量比对照提高了960μg·g-1。在大豆种子成熟过程中,籽粒中的苯丙氨酸解氨酶的活力呈低-高-低的单峰曲线;色氨酸呈由低-高的变化趋势;黄豆苷元,染料木黄酮的含量呈低-高-低-上升的趋势,总异黄酮的含量呈由低-高的变化趋势。Isoflavones play a vital role in health benefits,and the level is very low even in redundant soybean (about 3%).Increase the level in soybean is useful to improve quality of soybean.Exogenous methyl jasmonate were used as inducer,sprayed it on the leaf,to up-regulate the pathway of isoflavones synthesis.Ultraviolet spectrophotometer,fluorescent scanning (PE.USA) and HPLC (Agilent,Wilmington) were used to analyze the dynamic change of the activity ofisofalvones level.The results showed that exogenous methyl jasmonate induce the activity of PAL and the level of tryptophane,so that the synthesis of daidzein,genistein and total isoflavones increased.In the sprayed concentration scope of 0-450 μmol·L-1,450 μmol·L-1 was an appropriate concentration to induce the synthesis of isoflavones.The daidein level of the seeds treated with 450 μmol·L-1 increased by 5.1 μg·g-1,the genistein level of the seeds treated with 5.1 μg·g-1 and total isoflavones level increased by 960 μg·g-1.In the period of reproductive growth,PAL activity in seeds presented low-high-low single peak trend,daidzein and genistein present low-high-low-getting higher trend,tryptophan and total isoflavones present low-high-low trend.
