摘 要:拉曼光谱是一种简便灵敏的光谱分析新技术,近年来在细胞的结构、功能等方面的研究取得了迅速发展。本文系统的分析了药物曲古菌素A作用于宫颈癌细胞(HeLa细胞)24小时后的拉曼光谱变化,探讨了曲古菌素A作用后细胞内生物分子的结构及其含量的变化规律。结果表明:曲古菌素A作用后HeLa细胞中蛋白质的主链和侧链、核酸、脂类的谱带都发生了一定的改变,其中经200nM曲古菌素A处理后的样品谱带变化尤其明显,即拉曼光谱变化的规律与曲古菌素A的剂量存在一定的关系。该研究为曲占菌素A的临床应用奠定了基础,也为其它潜在抗癌药物的筛选提供了新思路。Raman spectra is a simple and sensitive technology.In recent years,Raman spectra have been rapid applied in cellular structure and function research.Now,the Raman spectra of HeLa cells having been treated by Trichostatin A for 24h were studied in this paper.Then,the change rule of molecule structure and content in cells were discussed.Results indicate the spectra band of protein or nucleic acid or lipid changed when HeLa cells were treated by Trichostatin A,and the spectra changes of cells were evident particularly after 200nM Trichostatin A treated,which indicated there was relationship between the change rule and the dose of Trichostatin A.This study prepares the ground for Trichostatin A to be applied in clinic,and supplies a new idea to screen other potential anticancer drugs.
