摘 要:[目的]研究微针注射甜玉米(Zea mays L.)胚芽gus基因转化瞬时表达,为玉米遗传转化的进一步研究奠定基础。[方法]以农杆菌为介导,采用gus基因瞬时表达技术,建立了胚芽注射开放性基因转化体系。[结果]3 mm长的胚芽为最适转化受体;农杆菌EHA105菌液浓度最适OD值为0.558~0.630;gus基因转化最适共培养天数为3 d。在以上最适转化条件下,微针注射胚芽生长点基因转化率为1%。[结论]该研究可为改良甜玉米遗传性状,培育基因工程新品种提供参考。[Objective] The aim to study the gus gene transformation transient expression on microneedle injection sweet corn germ,to lay basis for deeply researching corn transformation.[Method] The experiment was done by using gus gene transient expression technology mediated by agrobacterium,and a embryo injection system for open gene transformation was built.[Result] The results showed that the seeds germ with the length of 3 mm was the most optimal transformation receptor.The optimal OD value of EHA105 agrobacterium of bacterial concentration was 0.558-0.630.The optimal days of co-culture in gus gene transformation was 3 days.Under the optimal conditions,the microneedle injection of germ growth point gene conversion rate was 1%.[Conclusion] The study can provide reference for improving the hereditary character and cultivating new varieties of genetically engineering of sweet corn.
