摘 要:目的:观察远志清脑颗粒对拟痴呆模型小鼠学习记忆能力的影响及相关机制。方法:用ICR小鼠,设空白对照组,模型对照组,天保宁40 mg·kg-1组,远志清脑颗粒低、中、高剂量组(375,750,1 500 mg·kg-1),采用ip D-半乳糖(D-gal)合并亚硝酸钠(NaNO2)造痴呆模型小鼠。给药后通过水迷宫行为学实验测定模型小鼠的学习记忆能力,病理染色观察皮层和海马神经元的变化,分光光度法测定其脑组织生化代谢——超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力、丙二醛(MDA)含量。结果:远志清脑颗粒750 mg·kg-1组小鼠的学习能力和记忆能力与模型组相比显著提高(P<0.01);大脑SOD(1.64±0.26)U·mg-1较模型组(0.88±0.25)U·mg-1明显升高(P<0.01);GSH-Px(0.297±0.051)U·mg-1较模型组(0.179±0.035)U·mg-1升高(P<0.01);脑组织MDA(2.71±0.80)μmol·g-1较模型组(3.72±0.95)μmol·g-1显著降低(P<0.01)。结论:远志清脑颗粒明显提高小鼠的学习能力和记忆能力,并且可明显升高大脑SOD,GSH-Px活性,降低脑组织MDA含量,这可能是其改善学习能力和记忆能力的基础。Objective:To observe the influence of Yuanzhi Qingnao Keli(YZQNKL) on learning and memory ability and biochemical indexes in dementia model of mouse.Method:This research used ICR mice,injected D-gal and the nitric acid sodium lead to induce dementia in mice.Treatments were carried out with drugs.The maze was used to evaluate the learn ability of mice.The brain tissue biochemical indexes(SOD GSH-Px and MDA)were determined by spectrophotometry.Pathological dyeing and morphological examnation were carried out to investigate changes in cortex and hippocampal neurons.Result:YZQNKL of 750 mg·kg-1 improved learning ability singnificantly compared with that of the controls(P 0.01).YZQNKL of 750 mg·kg-1 could obviously increase the levels of SOD(1.64±0.26)U·mg-1,GSH-Px(0.297±0.051)U·mg-1(P0.01)and reduce MDA(2.71±0.80)μmol·g-1in brain tissue(P0.05).Conclusion:YZQNKL may improve learning and memory ability,and increase levels of SOD,GSH-Px,and reduce MDA in brain tissue.
