摘 要:目的研究白三烯与动脉粥样硬化的关系,探讨孟鲁司特对动脉粥样硬化的作用及意义。方法选取正常雄性SD大鼠75只,随机分为正常对照组、动脉粥样硬化组、孟鲁司特钠1mg组、孟鲁司特钠3mg组和辛伐他汀组。正常对照组持续普通饲料喂养,其余四组高脂饲料喂养,建立动脉粥样硬化模型后,孟鲁司特钠组、辛伐他汀组再分别给予孟鲁司特钠、辛伐他汀灌胃干预4周,18周末处死大鼠,采血检测血脂及血清白三烯B4(LTB4)、白三烯C4(LTC4)含量,分离腹主动脉,一部分经HE染色和透射电镜观察内膜病理表现,一部分制成主动脉匀浆检测白三烯B4、白三烯C4的含量。结果动脉粥样硬化组、孟鲁司特钠1mg组、孟鲁司特钠3mg组血脂较正常对照组明显升高(P<0.01);动脉粥样硬化组、孟鲁司特钠1mg组、辛伐他汀组白三烯B4、C4含量较正常对照组升高(P<0.05);孟鲁司特钠3mg组白三烯C4含量较动脉粥样硬化组降低(P<0.05)。动脉粥样硬化组内膜增生较明显,与其相比较,孟鲁司特钠1mg组无明显差异,而孟鲁司特钠3mg组和辛伐他汀组内膜厚度变薄。结论白三烯B4、白三烯C4参与动脉粥样硬化的损伤,孟鲁司特钠能降低白三烯C4含量,减轻动脉粥样硬化损伤,无降低血脂的作用。Objective To explore the relation betweet leukotriene and atherosclerosis and to demonstrate the effect of montelukast sodium on atherosclerosis. Methods Seventy - five normal male SD rats were divided into five groups randomly and they were normal control group, atherosclerotic group, montelukast sodium 1mg group,montelukast sodium 3mg group and simvastatin group, normal control group was fed with normal diet and the other groups were fed with high cholesterol diet in order to establish the model of atherosclerotic rats. After 14 weeks, montelukast sodium lmg group,montelukast sodium 3rag group and simvastatin group were treated with montelukast sodium and simvastatin for 4 weeks, then the rats were killed. Blood lipids and LTB4, LTC4 were measured in five groups, and aorta were observed by Hemotoxylin -Easin staining. Results Blood lipids in atherosclerotic group,montelukast sodium lmg group,montelukast sodium 3mg group was sig- nificantly higher than those in normal control group( P 〈 0.01 ) ; Blood lipids in simvastatin group was significantly lower than those in atherosclerotic group ( P 〈 0.01 ) ; Blood and aorta LTB4, LTCA in atherosclerotic group,montelukast sodium lmg group and simvastatin group was significantly higher than those in normal control group (P 〈 0.01 ) ;LTC4 in montelukast sodium 3 mg group was significantly lower than those in atherosclerotic group ( P 〈 0.01 ). Conclusion The expression of LTIM, LTC4 in blood and artor of atherosclerotic rats was abnormal. Montelukast sodium can reduce LTC4 and restrain incidence of atherosclerosis.
