摘 要:目的:研究氧化苦参碱(OMT)对大鼠冠脉结扎诱发实验性急性心肌梗死的保护作用。方法:采用结扎SD大鼠冠状动脉左前降支(LAD)6h复制大鼠实验性急性心肌梗死模型。血流动力学指标分析心功能;心电图J点位移和NBT染色法分析心肌梗死范围(坏死及缺血范围);比色法测定血清中乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)活性。结果:OMT 50mg/kg组能明显改善大鼠冠脉结扎后诱发急性心肌梗死心功能的紊乱;减轻结扎冠状动脉致心肌损伤范围、减小心肌梗死率;抑制心肌梗死大鼠心电图J点位移的升高;降低心肌梗死大鼠血清中CK、LDH活性。结论:OMT对大鼠冠状动脉结扎诱发实验性急性心肌梗死血流动力学具有显著的改善作用,对心肌梗死具有一定的保护作用。Objective: To investigate the protective effects of oxymatrine (OMT) on experimental acute myocardial infarction (AMI) in rats induced by left anterior descending branch of coronary artery. Methods: The AMI model was reproduced in rats by ligation of left anterior descending branch of coronary artery after 6 hours. The cardiac function was assayed by hemodynamic parameters; The myocardial infarction ratio and ischemic field were detected by NBT staining and change of J point shift on the ECG of postoperation; The activities of LDH and CK in serum were assayed by colorimetry with commercial kits. Results: Cardiac function was remarkable disfunction according to the results of the hemodynamics, myocardial infarction ratio and J point shift were significantly increase, and LDH and CK activities in serum were significantly increase as well. OMT remarkable ameliorated the cardiac function, decreased myocardial infarction ratio and J point shift, and inhibited the LDH and CK activities in serum. Conclusion: OMT ameliorate exacerbating cardiac function and protect acute myocardial infarction in rats induced by ligating left anterior descending branch of coronary artery.
