摘  要:目的:研究乙醇对钾通道阻断剂四乙胺(Tetraethylammonium,TEA)对大鼠大脑中动脉自律性收缩作用的影响及机制。方法:大鼠大脑中动脉环固定在DMT多道记录系统浴槽内传感器上,通过Chart 5.4记录TEA作用下血管环张力的变化,并观察各种工具药对TEA缩血管作用及自律性的影响。结果:TEA(10-60mmol·L^(-1))可引起大脑中动脉浓度依赖性收缩,并产生自律波。TEA类似物gallamine(2-10mmol·L^(-1))引起浓度依赖性收缩,但不引起自律波。钙拮抗剂普萘洛尔(1μmol·L^(-1)),硝苯地平(1μmol·L^(-1))和LaCl_3(1μmol·L^(-1))均明显抑制TEA的收缩血管的作用,并且取消了TEA产生的自律波。α肾上腺素能受体阻断剂哌唑嗪(10μmol·L^(-1)),育亨宾(10μmol·L^(-1))和血管紧张素口受体拮抗剂氯沙坦(1μmol·L^(-1))对TEA的缩血管作用及自律波均无明显影响。血管紧张素转换酶抑制剂依那普利(0.1mmol·L^(-1))抑制了TEA的缩血管作用,但对TEA产生的自律波无作用;1%乙醇明显增强了TEA的缩血管作用,并且完全抑制了TEA产生的自律波。结论:TEA缩血管作用所产生的自律波可能与TEA的结构,胞浆内Ca^(2+)度增加有关;乙醇可能通过阻滞血管平滑肌上的某些钾通道而取消其自律性。Objective:To investigate the vasoconstrictive roles,autorhythmicity and the possible mechanisms of tetraethtylamine (TEA) on middle cerebral artery of rats.Methods:Rat isolated middle cerebral arteries were mounted on a wire myograph and the tensions were recorded.The vasoconstrictive action of TEA,antorhythmicity and the influence of various drugs on it were observed in the rings with endothelium.Results:TEA (10-60 mmol.L^-1) caused concentration-dependent contraction and autorhythmic waves in middle cerebral arteries with endothelium;gallamine (2-12 mmol.L^-1),a trivalent TEA analog,caused concentration-dependent contraction, but not autorhythmic waves. Propranolol (1 μmol.L^-1),nifedipine (1μmol-L^-1) and LaCI3(1 μmoI-L^-1) inhibited the contraction and abolished the autorhythmic waves induced by TEA (20 mmol.L^-1)in middle cerebral artery rings. Prazosin (10μmol.L^-1), yohimhine (10μmoL.L^-1) and Iosartan (lμmol.L^-1) had no influnce on the contraction and autorhythmic waves in rings;enalapril (0.1mmol.L^-1) inhibited the contraction, but had no influnce on the autorhythmic waves induced by TEA (20 mmol.L^-1) in rings. 1% alcohol potentiated the TEA-induced contraction and abolished the autorhythmic waves in middle cerebral artery rings.Conclusion:The vasoconstrictive action of TEA is associated with the increase of Ca^2+ in cytoplasm;the structure of TEA,concentration of Ca^2+ in cytoplasm and alcohol may be involved in the action of TEA.
