摘 要:目的:建立测定人体血浆中多潘立酮的高效液相色谱(HPLC)法,并测定健康志愿者口服多潘立酮片24h内血药浓度。方法:血浆样品碱化后以有机溶剂提取,色谱柱为Inertsil ODS-35μm 150×4.6mm,0.05moL·L^(-1)磷酸二氢钠(pH2.8):乙腈:四氢呋喃(78:22:0.5)为流动相,流速1.4ml·min^(-1),荧光检测波长为λex=285nm,λem=325rm。结果:多潘立酮本法线性范围为:1~100ng·ml^(-1)(r=0.9995,n=7);最低定量限1ng·ml^(-1);低、中、高3种浓度的平均回收率分别为(109.48±3.90)%,(108.5±1.50)%,(102.76±0.32)%;日内、日间精密度RSD均<4%。结论:本检测方法简便、快速、经济、准确,可用于测定人血浆中多潘立酮的浓度。Objective:To establish an HPLC method for the determination of domperidone in human plasma, and to determine the concentration of domperidone in human plasma after oral of domperidone tablets into healthy volunteers within 24 hours.Methods:The domperidone was extracted from plasma by organic solvents.Chromatographic separation has been achieved on Inertsil ODS-35pm (150×4.6mm) with 0.05moL.L^-1 sodium dihydrogen phosphate(pH2.8):acetonitrile:tetrahydrofuran (78:22:0.5) as themobile phase at a flow rate of 1.4ml.min^-1; The fluorescence detection atλex=285nm and λem=325nm.Results:The linear range wasl-100ng-ml^-1 (r=0.9995,n=7).The minimum ration limit was lng.ml^-1;The average recoveries of low, middle and high concentration were (109.48±3.90)%,(108.5±l.50)% and (102.76±0.32)%, RSDs of inter and intra-day preeisions were legs than 4%.Conclusion:The method is simple, rapid, economic and accurate for determination of domperidone in human plasma.
