摘 要:目的:观察通心络对动脉粥样硬化(AS)模型兔基质金属蛋白酶MMP-2,MMP-9(简称MMP-2,9)和其天然抑制物(TIMP-1)介导作用的影响及其可能机制。方法:运用球囊损伤腹主动脉加高脂饮食建立兔AS模型。38只新西兰兔随机分成5组:空白组(6只)、模型组(8只)、通心络组(8只)、辛伐他汀组(8只)和通心络加辛伐他汀组(8只)。给药8周后采集耳缘静脉血检测血脂;多普勒检测髂动脉和主动脉的血流速度;免疫组织化学方法检测MMP-2,9、TIMP-1的光密度值。结果:与模型组比较,通心络组可减轻AS病变;降低血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)水平(P<0.05);提高血流速度(P<0.05);下调MMP-2,9的表达(P<0.05);提高TIMP-1的表达;通心络合用辛伐他汀比单用辛伐他汀能进一步减少斑块的数目和降低MMP-2,9的表达。结论:通心络有明显降低血清总胆固醇的作用,同时其有加快血流速度的作用,此作用可能与其抑制AS模型兔血管壁中MMP-2,9及提高TIMP-1的表达,稳定斑块有关。Objective:To observe Tongxinluo(TXL) on atherosclerosis(AS) of rabbit matrix metalloproteinase(MMP-2,9)and its natural inhibitor TIMP-mediated effect possible mechanism.Methods:Balloon catheter denuding injury abdomen aorta and high fat diet were performed to establish rabbit AS model.New Zealand rabbits were randomly divided into 5 groups: the normal group(n=6),the model group(n=8),the TXL treated group(n=8),the Simvastatin treated group(n=8) and the TXL+ Simvastatin treated group(n=8).The rabbits were killed after 8 weeks,serum lipid levels were examined respectively.Doppler examined iliac artery and aortic blood flow velocity;immunohistochemistry detected MMP-2,9,TIMP-1 optical density value.Results:Compared with model group,TXL group can lessen AS disease,lower serum total cholesterol,triglyceride,low density lipoprotein levels(P0.05),increase blood flow velocity(P0.05),reduce MMP-2,9 expression(P0.05).enhance the expression of TIMP-1.Both TXL and simvastatin complex can further reduce the number of blocks and the expression of MMP-2,9 than use simvastatin only.Conclusion:TXL lower serum total cholesterol significantly,and can speed up the blood flow velocity.This effect may be related to the inhibition of vascular wall in the AS model of MMP-2,9 and enhance the expression of TIMP-1 to stabilize plaque.
