摘 要:目的探讨转录因子T-bet/GATA3在过敏性哮喘患者免疫失衡中的作用。方法通过实时荧光定量PCR检测47例过敏性哮喘患者外周血单个核细胞(PBMC)中T-bet/GATA3 mRNA表达情况,用流式细胞仪检测其Th1、Th2细胞数量,以20名健康者为对照。结果过敏性哮喘患者外周血淋巴细胞转录因子T-bet mRNA表达水平低于正常对照组,且Ⅲ、Ⅳ级低于Ⅰ、Ⅱ级哮喘患者(P<0.05);转录因子GATA-3 mRNA表达水平高于正常对照组,以Ⅲ、Ⅳ级更高(P<0.05)。过敏性哮喘患者外周血淋巴细胞亚群Th1百分率低于正常对照组,其中Ⅲ、Ⅳ级低于Ⅰ、Ⅱ级哮喘患者(P<0.05);Th2细胞百分率显著高于正常对照组,Ⅲ级中度持续组与Ⅳ级重度持续组各哮喘组均显著高于Ⅰ级间歇发作组和Ⅱ级轻度持续组(P<0.05)。T-bet/GATA3与Th1/Th2结果一致。结论过敏性哮喘患者T-bet/GATA3 mRNA表达水平与Th1/Th2比例变化一致,似乎可以成为判断其细胞免疫能力的新指标。Objective To explore the effects of T-bet and GATA3 on profiles of type 1 and type 2 Th cells in allergic asthmatic patients. Methods To detect the mRNA expression of T-bet and GATA-3 in the PBMC by realtime quantitative PCR and determine Th1 and Th2 cell numbers by FACS in 47 mite asthmatic patients and 20 healthy control group.Results T-bet mRNA in allergic asthmatic patients was lower than those in control group,lower in Ⅲ and Ⅳ attack stage than in Ⅰ andⅡ remission stage(P0.05).GATA-3 mRNA was higher in mite asthmatic patients than control group and higher in Ⅲ and Ⅳ attack stage than in Ⅰ andⅡ remission stage(P0.05).Th1 percentage was lower in mite asthmatic patients than those in control group and lower in attack stage than those of remission stage(P0.05).Th2 percentage in mite asthmatic patients was higher than that in control group and higher in attack stage than that of remission stage(P0.05).The T-bet /GATA-3 mRNA ratio was coincidence with Th1 /Th2 ratio.Conclusion The T-bet /GATA3 ratio may act as a new measure of the Th1 /Th2 balance in mite asthmatic patients.
