摘  要:研究吸入放射性气体氡及其子体对大鼠肺组织的DNA损伤效应。采用雄性Wistar大鼠16只,随机分为4组,每组4只。动物整体暴露于多功能生态氡室,吸入氡及其子体的累积剂量分别达到64、121和236工作水平月(Working level month,WLM)。激光共聚焦检测肺泡灌洗液(BALF)细胞内活性氧(Reactive oxygenspecies,ROS)水平,ELISA法检测肺组织8-羟基脱氧鸟苷(8-hydroxydeoxyguanosine,8-OHdG)含量、RT-PCR检测肺组织8羟基鸟嘌呤DNA糖苷酶1型(8-oxoguanine DNA-glycosylase type 1,OGG1)mRNA和8-羟基脱氧三磷酸鸟嘌呤核苷酶(Nucleoside diphosphate linked moiety X-type motif 1,MTH1)mRNA相对表达水平。氡吸入染毒后肺组织8-OHdG含量显著升高,BALF细胞ROS增加,OGG1和MTH1表达呈下降趋势。氡及其子体吸入可导致肺组织细胞的DNA氧化损伤,主要表现为8-OHdG含量的增加和修复酶活性的降低。In order to study the effects on DNA damage in rat lung tissue after exposed to radon and its progeny, 16 male rats divided into 4 groups randomly were exposed to radon and its progeny at the cumulative doses of 0, 64, 121 and 236 working level month(WLM), respectively. The levels of ROS (Reactive oxygen species) in BALF (Bronchoalveolar lavage fluid) cells were determined by laser scanning confocal microscope meanwhile the content of 8-OHdG (8-Hydroxydeoxyguanosine) in lung tissue was detected by ELISA. The relative expression levels of OGG1 (8-oxoguanine DNA-glycosylase type 1) mRNA and MTH1 (Nucleoside diphosphate linked moiety X-type motif 1) mRNA were determined by RT-PCR. It was found that the ROS level and 8-OHdG content significantly increased, and the expression level of OGG1 decreased at the same time. The DNA damage of lung tissue exposed to radon and its progeny might be attributed to the increase of 8-OHdG level and the down-regulated level of relative DNA repairing enzymes.
