赤芍801对白细胞介素-6与其受体相互作用影响的研究

摘　　要：白细胞介素-6的可溶性受体(sIL-6R)在石英晶体微天平基片表面固定的量为297.08ng.cm-2。白细胞介素-6(IL-6)可以特异性结合sIL-6R,结合的量为33.70 ng.cm-2。IL-6的响应具有计量依赖性。根据上述分析,得到kass值为1.27×104(mol/L)-1s-1,kdiss值为1.94×10-4s-1,KD值为1.53×10-8mol/L。表明赤芍801能够增加IL-6与其受体的结合,从而影响IL-6及其受体的生物学活性。可以推断出IL-6与其受体的结合是赤芍801在体内的另一个作用靶点。The immobilized amounts of interleukin-6 soluble receptor（sIL-6R） on the surface of quartz crystal microbalance（QCM） substrate are 297.08 ng·cm-2.interleukin-6（IL-6） can specifically bind to sIL-6R and the bound amounts are 33.70 ng·cm-2.The response for IL-6 is dose-dependent.Based on the analysis,the value for kass is 1.27×104（mol/L）-1s-1,the value for kdiss is 1.94×10-4s-1,the value for KD is 1.53×10-8 mol/L.Paeoniae radix 801 can enhance the binding amounts of IL-6/sIL-6R,which is dose-dependent.IL-6/sIL-6R is another action target of Paeoniae Radix 801 in vivo.
