摘  要:目的观察自由基清除剂依达拉奉对急性一氧化碳中毒性脑病大鼠空间记忆及海马CA1区神经元凋亡的影响。方法雄性SD大鼠40只,随机均分为正常组、急性一氧化碳中毒性脑病(TE-ACMP)组、生理盐水组、依达拉奉组4组。采用Morris水迷宫检测4组SD大鼠的空间记忆,以平均逃避潜伏期衡量空间记忆能力;TUNEL法检测神经元海马CA1区神经元凋亡情况。结果 ACMP组较正常组大鼠平均逃避潜伏期明显延长(P<0.05),而依达拉奉组大鼠平均逃避潜伏期较TE-ACMP组和生理盐水组均显著缩短(P<0.05)。与正常组相比,TE-ACMP组海马CA1区凋亡指数明显升高(P<0.01)。与TE-ACMP组、生理盐水组相比,依达拉奉组海马CA1区凋亡指数明显降低(P<0.01)。结论依达拉奉可改善TE-ACMP大鼠受损的空间记忆能力,抑制TE-ACMP大鼠海马CA1区神经元凋亡,可用于临床防治一氧化碳中毒性脑病。Objectives To explore the effect of edaravone on the spatial memory and the apoptosis of hippocampal CA1 area in rats with toxic encephalopathy after acute carbon monoxide poisoning(TE-ACMP).Methods Forty male SD rats were randomly divided into four groups,i.e.control group,TE-ACMP group,normal saline group and edaravone group.The spatial memory was examined by Morris water maze test,apoptosis was measured by TUNEL method.Results The average latency of Morris water maze test in TE-ACMP group was significantly longer than that in control group(P〈0.05),the average latency in edaravone group was significantly shorter than that both in saline group or in TE-ACMP group(P〈0.05).Meanwhile,the apoptosis index in TE-ACMP group was significantly higher than that in control group(P〈0.01),whereas,the apoptosis index in edaravone group was significantly lower than that in saline group(P〈0.01).Conclusions Edaravone might ameliorate the damaged spatial memory and apoptosis in hippocampus area of TE-ACMP rats,suggesting it is helpful for the clinical management of acute CO poisoning.
