摘 要:目的探讨培哚普利对自发性高血压大鼠(SHR)肾脏水通道蛋白2(AQP2)表达和尿液AQP2排泄的影响。方法 SHR20只,随机分成2组,每组10只,分别为对照组、培哚普利治疗组,经用药后观察血压、血Na+、尿量以及尿渗透压,用放射免疫法检测血浆血管加压素(AVP)浓度,用免疫组化、RT-PCR、Western blot检测肾脏AQP2表达,用间接ELISA法检测尿液AQP2浓度。结果 SHR经培哚普利治疗8周后,血压下降(P<0·05)、血Na+水平差异无统计学意义,24h尿量增多(P<0·05)、尿渗透压低于对照组(P<0·05)。免疫组化结果显示,培哚普利治疗后肾组织AQP2表达明显减弱(P<0·05);用RT-PCR和Western blot方法检测表明,与对照组比较,治疗组肾组织AQP2 mRNA和蛋白的表达均明显减少(均P<0·05);血浆AVP浓度低于对照组(P<0·05)、尿液AQP2浓度高于对照组(P<0·05)。结论培哚普利下调SHR肾脏组织的AQP2 mRNA和蛋白表达,增加AQP2尿液排泄。Objective To investigate the effects of perindopril on renal aquaporin-2(AQP2) expression and urine aquaporin-2(U-AQP2) excretion in spontaneously hypertensive rats (SHRs). Methods Twenty SHRs were randomized into control group(SHRctrl) and perindopril group (SHRper) (n=10 per group). Blood and urine samples were collected from the SHR for detecting blood presssure, serum Na+, urine volume and urine osmolality after perindopril treatment. Plasma arginine vasopressin (p-AVP) concentration was detected by radioimmunoassay (RIA). Immuno-histochemistry, semi-quantitative real time-polymerase chain reaction (RT-PCR) and Western-blot were performed to measure the expression of kidney AQP2. U-AQP2 concentration was detected by indirect enzyme-linked immunosorbent assay (ELISA). Results After 8 weeks, blood pressure and 24 hours urine osmolality were lower in SHRper than in SHRctrl(P0.05), and 24-hour urine volume increased in SHRper(P0.05), while serum Na+ did not differ significantly between the two groups(P0.05). Immuno-histochemistry analysis showed that the treatment resulted in marked down-regulation on renal AQP2 expression in SHRper(P0.05). RT-PCR and Western blot also revealed significantly decreased levels of kidney AQP2 mRNA and AQP2 protein in SHRper(P0.05, respectively). Compared with those in SHRctrl, p-AVP concentration was lower in SHRper(P0.05), whereas U-AQP2 concentration was higher in SHRper (P0.05). Conclusion Perindopril can reduce the expression of AQP2 in the kidney inner medullary, and accelerate the excretion of U-AQP2 in SHRs.
