摘 要:目的探讨大鼠闭合性颅脑损伤后脑组织微管相关蛋白-2(MAP-2)表达变化规律。方法建立大鼠闭合性颅脑挫伤模型6,4只Wistar大鼠随机分为损伤后0.5h、6h1、2h、1d3、d7、d1、4d7个损伤组及对照组,每组8只。应用免疫组化和蛋白印记Western blot法及图像分析技术,检测脑挫伤后各设定时间点挫伤脑组织中MAP-2的变化。结果免疫组化染色显示:对照组大鼠脑组织中可见MAP-2阳性表达;实验组伤后0.5h,挫伤灶及周围MAP-2阳性表达下降,伤后6h阳性表达降至最低,伤后12h,阳性表达逐渐增多,伤后14d,仍保持较高水平;蛋白印记结果与免疫组化结果一致。统计学分析显示:MAP-2的表达各损伤组与对照组比较,差异均有统计学意义(P<0.05),伤后12h、3d及14d与上一组比较,差异有统计学意义(P<0.05)。结论大鼠脑挫伤后14d内,MAP-2在脑组织挫伤灶及其周围呈现先减少后逐渐增多的表达变化,可作为推断脑挫伤经过时间的参考指标之一。Objective To investigate the expression of MAP-2 after brain contusion and its significance for estimating the age of brain contusion. Methods Sixty-four rats were randomly divided into eight groups:7 injury groups of 0.5h ,6h, 12h, l d ,3d,7d, 14d post-injury respectively and a control group. The expression of MAP-2 at different time was detected by immunohistoehemical stainir^g and Western blot. Results The positive staining of MAP-2 appeared in the control group,but declined gradually in 0. 511 after contusion in the injury group and reached valley bottom in 6 hours after contusion, then increased gradually in 12 hours after contusion,and could be observed at a higher level in 14 days after contusion. The results of western blotting were consistent with the immunohistoehemical staining. Statistical analysis showed: The differences of the MAP-2 expression is of statistically significant (P 〈 0.05) between every injury group and the control group, and between anyone of the three groups of 12 hours after injury,3d and 14d after injury, and its previous group. Conclusion Within 14 days after brain contusion,the time-dependent changes of MAP-2 expression may be used to the dating of wound of brain contusion in forensic practice.
