摘  要:目的:研究肺表皮生长因子受体(EGFR)表达与肺纤维化发生的关系。方法:将54只Wistar大鼠随机分成模型组、易瑞沙(iressa)组和对照组,模型组和iressa组使用博莱霉素(5 mg.kg-1)气管注射制造鼠肺纤维化模型,对照组气管注入生理盐水0.2~0.3 ml。iressa组、模型组和对照组造模前1 h分别灌胃给予iressa(200 mg.kg-1)、生理盐水10 m.lkg-1和生理盐水10 ml.kg-1,5次.周-1,分别于第1、14、28天处死大鼠。用SP免疫组化法研究原癌基因和肺EGFR在不同程度肺纤维化条件下的表达。结果:(1)模型组和iressa组EGFR表达在各时间点与对照组差异有统计学意义;(2)模型组EGFR表达在第28天达到顶峰,此时间点EGFR表达与同组其他时间点差异均有统计学意义;(3)iressa组EGFR水平下降,同时较模型组肺纤维化程度减轻(P<0.01)。结论:EGFR在肺纤维化不同阶段表达增强,提示EGFR可能参与肺纤维化发生过程,且可被其拮抗剂影响。Objective: To study the effects of iressa on epidermal growth factor receptor(EGFR) expression at the different time points on the pulmonary fibrosis induced by Bleomycin in rats.Methods: Fifty-four Wistar rats were randomly divided into three groups,the model group,iressa group and the control group.They were sacrificed on days 1,14 and 28 after operation,respectively.Samples were harvested for further studies.Results:(1) Compared with the control group,the expressions of EGFR were increased significantly in the model group and the iressa group.(2) The expressional level of EGFR arrived at a peak at the 28th day in the model group.(3) After treatment with iressa the expressions of EGFR were decreased significantly at the 14th and 28th day(P0.01).Likewise the fibrosis was alleviated significantly.Conclusion: The expressions of EGFR are dynamically increased during the development of the experimental pulmonary fibrosis,which could be affected by the EGFR antagonists iresssa.These support the EGFR might play a important role in the development of pulmonary fibrosis.
