摘  要:目的观察人体退变椎间盘组织中血小板源性生长因子-A(PDGF-A)、血小板源性生长因子受体-α(PDGFR-α)的表达,探讨其生物学意义。方法免疫组化(S-P)法分别检测PDGF-A、PDGFR-α、增殖细胞核抗原(PCNA)在正常对照组(A组,n=17)、退变椎间盘组(B组,n=51)中的表达;TUNEL法检测两组椎间盘细胞凋亡情况。结果 PDGF-A、PDGFR-α在B组中表达低于A组,两组之间差异有统计学意义(P<0.05);细胞增殖指数(PI)在B组数值低于A组,细胞凋亡指数(AI)在B组数值高于A组,两组之间差异有统计学意义(P<0.05);PDGF-A、PDGFR-α的表达与PI正相关、与AI负相关(P<0.05);PI、AI值在PDGF-A、PDGFR-α阴性和阳性组中差异有统计学意义(P<0.05)。结论 PDGF-A、PDGFR-α的异常表达与人体椎间盘退变密切相关;PDGF-A、PDGFR-α促进细胞增殖,抑制细胞凋亡,在人体椎间盘退变过程中起了重要的作用,其异常表达可能导致椎间盘退变的发生。Objective To observe the expression of PDGF-A and PDGFR-α in human degenerated intervertebral discs and investigate its biological significance.Methods 68 patients were divided into two groups:17 cases of normal intervertebral discs(group A),51 cases of degenerated intervertebral discs(group B).The expression of PDGF-A、PDGFR-α,PCNA and apoptosis were detected by immunohistochemical and TUNEL.Results The expression rate of PDGF-A and PDGFR-α in group B was lower than group A,the difference was significant between them(P0.05).PI in group B was significant lower than that in group A(P0.05).AI in group B was significant higher than that in group A(P0.05).The expression of PDGF-A and PDGFR-α had positive correlation with PI and negative correlation with AI(P0.05).There was significant difference in PI and AI between positive expression group and negative expression group(P0.05).Conclusion Abnormal expression of PDGF-A、PDGFR-α is closely related to degenerative intervertebral discs;PDGF-A and PDGFR-α promote cell proliferation and inhibit intervertebral discs cell apoptosis,which has an important effect on the degeneration of intervertebral discs.Abnormal expression of PDGF-A、PDGFR-α can induce degeneration of intervertebral discs.
