摘 要:目的:探讨低氧暴露后大鼠外周血T淋巴细胞亚群及活化共刺激分子的变化,为干预措施的研究提供科学依据。方法:采用三色免疫荧光标记流式细胞仪分析法,观察在低氧暴露前和模拟海拔8 000 m低氧暴露8 h、3d6、d和10 d后,大鼠外周血T淋巴细胞亚群和T淋巴细胞活化共刺激分子的变化。结果:模拟海拔8 000 m低氧暴露8 h后,大鼠CD3+、CD8+、CD8+CD28-细胞数较低氧前均显著降低(P均<0.01);低氧暴露3 d后,除上述变化外,CD4+CD28+细胞数也显著降低(P<0.01),CD4+CD28-细胞数显著增加(P<0.01);低氧暴露6 d和10 d后,CD3+、CD4+呈现进一步降低趋势,CD8+CD28+细胞数显著增加(P<0.01)。结论:说明模拟海拔8 000 m低氧暴露8 h和3 d后,大鼠外周血CD8+、CD4+T淋巴细胞活化水平均显著下降,随着低氧暴露时间的延长,CD8+T淋巴细胞活化水平由降低变为增加。Objective:To explore the effect of hypoxia on the peripheral blood T lymphocyte subsets and co-stimulatory molecules in rats so as to provide the basis for studying the intervention measure.Methods:Before hypoxia and during hypoxia at 8 000 m for 8 h,3 d,6 d and 10 d the change of peripheral blood T lymphocyte subsets and co-stimulatory molecules in rats were detected by flowcytometer with three-color immunofluorescence label.Results:Rats were exposed to hypoxia at 8 000 m for 8 hours,and CD3+,CD8+,CD8+CD28-lymphocyte percentages were significantly decreased(P0.01) compared with that before hypoxia.After 3 days of hypoxia,besides aforesaid change,CD4+CD28+ lymphocyte percentage also prominently decreased(P0.01) and CD4+CD28-prominently increased(P0.01).After 6 and 10 days of hypoxia,CD3+、CD4+ lymphocyte percentages were further decreased,while CD8+CD28+ lymphocyte percentage significantly increased(P0.01).Conclusion:After exposed to hypoxia at 8 000 m for 8 hours and 3 days,activation of CD8+ and CD4+ T lymphocyte was prominently decreased,while with the prolong of exposed time activation of CD8+ T lymphocyte was significantly increased.
